The ingredients involved — glucosamine hydrochloride, chondroitin sulfate, sometimes MSM and manganese — have a long safety record in dogs at label amounts. The genuinely serious problems documented in the veterinary literature involve massive accidental ingestion, not daily use, and they trace back to manganese rather than to glucosamine.

Quick answer: Reported side effects of Cosequin in dogs are mainly gastrointestinal — loose stool, vomiting, reduced appetite, gas — and they tend to appear early and resolve. Allergic-type reactions (facial swelling, hives, itching) are described but rare. The one well-documented severe scenario in the veterinary literature is acute liver failure after a dog eats a large quantity of joint supplement tablets at once, linked to manganese toxicosis rather than to glucosamine or chondroitin. What is actually in Cosequin, and why does that matter for side effects?

You can’t reason about side effects without knowing what’s in the tablet. Nutramax publishes the active ingredient panel on its own product pages. For Cosequin Chewable Tablets with MSM, the label states each chewable tablet contains glucosamine hydrochloride (FCHG49) 600 mg, sodium chondroitin sulfate (TRH122) 300 mg, methylsulfonylmethane (MSM) 250 mg, and manganese (as manganese ascorbate) 3 mg.

That fourth line is the one owners skip and the one that matters most in an overdose. Manganese is an essential trace mineral at small amounts and a hepatotoxin at large ones. Everything else on the panel has a wide margin of safety; manganese is the ingredient that turns a chewed-through bottle into an emergency.

What side effects are reported in dogs, and how strong is the evidence?

Here’s the honest picture, sorted by body system and by how much evidence actually stands behind each item. Nothing in this table is a diagnosis, and none of it predicts what your individual dog will do.

In the randomized, double-blind, positive-controlled trial by McCarthy and colleagues (The Veterinary Journal, 2007), 35 dogs with confirmed hip or elbow osteoarthritis received either a glucosamine hydrochloride/chondroitin sulfate combination or carprofen as a positive control. The glucosamine/chondroitin group showed statistically significant improvement in pain, weight-bearing and severity scores by day 70, with a slower onset than the carprofen group. Safety, not efficacy, is the point here: the trial ran ten weeks of daily dosing in client-owned dogs without a safety signal emerging as the story of the paper.

A later and less flattering trial is worth knowing about for the sake of expectation-setting. Kampa and colleagues (Frontiers in Veterinary Science, 2023) randomized 75 owned dogs with hip osteoarthritis across five groups including glucosamine/chondroitin, two marine fatty acid compounds, carprofen and placebo, using force-plate peak vertical force as the objective outcome. At four and six weeks, the marine compounds and carprofen improved significantly over placebo; the glucosamine/chondroitin group did not. Bhathal and colleagues, reviewing the canine literature in Open Veterinary Journal (2017), reached a similar verdict — evidence limited and conflicting, with a shortage of well-designed trials.

That combination — a benign side effect profile alongside genuinely mixed efficacy evidence — is the fair summary. The risk of trying it is low. How dangerous is an accidental overdose?

This is the part of the topic that deserves more attention than it usually gets, because it’s where the documented harm lives.

ASPCA Pro’s toxicology guidance notes that oral chondroprotective agents have historically been considered to have wide safety margins, citing oral glucosamine LD50 figures in animals around 8,000 mg/kg with no adverse effects reported at 2,700 mg/kg over 12 months — figures from the toxicology literature, not dosing advice for you. Their guidance also flags the shift in thinking: a published case of hepatic failure after a joint supplement overdose, with severely elevated manganese in liver and kidney.

That case is Borchers and colleagues (Journal of Veterinary Diagnostic Investigation, 2014), from UC Davis. A 5-year-old spayed female Pug presented with vomiting and ataxia after eating approximately 100 joint supplement tablets, developed acute hepatic failure, and was euthanized six days after presentation; necropsy showed severe acute hepatic necrosis with severely elevated manganese in liver and kidney tissue.

Jaffey and colleagues (Topics in Companion Animal Medicine, 2024) described two dogs in the same household that ate roughly 75 capsules of a human joint supplement, with 2,062 mg of manganese ingested between them. One developed fulminant liver failure with coagulopathy and died within 36 to 48 hours. The second developed severe acute liver injury, was hospitalized for six days with supportive care, and had clinical and biochemical resolution by day 44.

Three dogs across those two reports, one mechanism. This is a small literature by design — Jaffey and colleagues note that before their own paper, the published veterinary record of acute manganese intoxication in dogs consisted of two reports covering three dogs. The practical lesson is unglamorous: a palatable chewable your dog likes is a food item to him, and the bottle belongs behind a closed door. If your dog gets into the container, this is an emergency call to your veterinarian or a pet poison control line — not a search-and-monitor situation.

Call before you start if your dog is diabetic or has any known problem with glucose regulation, has existing liver or kidney disease, is on an anticoagulant or is scheduled for surgery, is pregnant or nursing, or is already taking another joint product — stacking two supplements can double an ingredient you never intended to double, manganese included.

Call during use for any reaction in the table above, for anything that starts within hours of a dose and repeats, or for a dog on long-term NSAIDs whose appetite or stool changes, since a supplement side effect and an NSAID side effect can look identical from the outside.

Call after any accidental ingestion of more than the labeled amount, with the bottle available so the active ingredient panel can be read verbatim.

Myth vs fact

Myth: my dog is allergic to shellfish, so glucosamine is dangerous for him. The glucosamine used in these products is typically derived from crustacean shell chitin, and shellfish allergy in people is usually a reaction to muscle protein rather than to shell material. Gray and colleagues examined this question directly in humans (Journal of Allergy and Clinical Immunology, 2004) and the concern has generally not held up. Canine-specific data are thin, so raise it with your vet rather than assuming either way.

Myth: a bigger dose works faster. The label pattern Nutramax publishes actually runs the other direction — a higher number of tablets for an initial period, then a lower maintenance number after that. Follow your product label or your veterinarian, and never scale by eye.

Myth: side effects show up months in. The reported pattern is early and digestive. A new problem appearing after a year of uneventful use is more likely to be something else, and deserves a workup rather than an assumption.

Myth: “natural” means no interactions. Look at a related joint product with an FDA-approved label: Adequan Canine, an injectable polysulfated glycosaminoglycan, states on its label that PSGAG is a synthetic heparinoid and is not to be used in dogs with known or suspected bleeding disorders. That’s a different product class from an oral chewable, and none of it transfers directly to Cosequin — but it illustrates why “supplement” doesn’t mean “tell no one.”

Can Cosequin cause diarrhea in dogs?
Loose stool is the most commonly described reaction, typically in the first days of use, and it usually resolves. Controlled trials report adverse events in dogs as uncommon overall. If diarrhea lasts more than a day or your dog is also vomiting or off food, pause and call your veterinarian.

Can Cosequin damage a dog’s liver?
Not at labeled amounts, based on published evidence. Acute liver failure has been documented in dogs after massive accidental ingestion of joint supplement tablets, and in those cases the liver injury was attributed to manganese toxicosis, with severely elevated manganese in liver and kidney tissue.
